金水鲜联合放疗对A549细胞凋亡及HIF—1α、VEGF表达的影响

[摘要] 目的 观察金水鲜联合放疗对肺腺癌A549细胞缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)表达的影响,并对其可能的机制进行探讨。 方法 常规培养细胞,随机分为四组:①对照组(NC组),加入50 μL生理盐水;②放疗组(RT组),单次照射,剂量2 Gy;③金水鲜组(JSX组),加入50 μL金水鲜;④金水鲜联合放疗组(JSX+RT组),加入50 μL金水鲜后对细胞进行单次放射线照射,剂量为2 Gy。CCK8测量实验组细胞增殖抑制率,酶联免疫吸附测定(ELISA)及Real Time-PCR检测各组HIF-1α、VEGF的表达情况。 结果 CCK8结果显示,JSX+RT组增殖抑制最明显(P < 0.05)。ELISA结果显示,在24 h时JSX+RT组HIF-1α、VEGF蛋白表达量分别为(284.27±2.66)、(462.40±3.58)pg/mL。RT-PCR结果显示,与其他各组比较,JSX+RT组HIF-1α、VEGF表达量明显下降(P < 0.05)。 结论 金水鲜联合放疗具有抑制肿瘤细胞增殖及HIF-1α、VEGF表达的能力。

Effect of Jinshuixian combined with radiotherapy for the apoptosis of A549 cells and the expression of HIF-1α and VEGF

[Abstract] Objective To observe the effect of Jinshuixian combined with radiotherapy on the expression of hypoxia inducible factor-1 alpha (HIF-1α) and vascular endothelial growth factor (VEGF) in A549 cells of lung adenocarcinoma, and to explore its possible mechanisms. Methods The cells were cultured generally, and they were randomly divided four groups: ①control group (NC group) was added with 50 μL saline; ②radiotherapy group (RT group) was given single irradiation, the radiation dose was 2 Gy; ③Jinshuixian group (JSX group) was added with 50 μL Jinshuixian; ④Jinshuixian + radiotherapy group (JSX+RT group), the cells were irradiated with a single radiation dose of 2 Gy after added 50 μL Jinshuixian. The CCK8 assay was used to measure the proliferation inhibitory rates of experimental groups, enzyme-linked immunosorbent assay (ELISA) and real time-PCR were introduced to test the expression of HIF-1α, VEGF. Results CCK8 test showed that the capable of inhibiting proliferation of JSX+RT group was the highest (P < 0.05). ELISA experiment showed that the expression of HIF-1α, VEGF in JSX+RT group were (284.27±2.66), (462.40±3.58) pg/mL in 24 h. RT-PCR experiment revealed that the expression of HIF-1α, VEGF in JSX+RT group were obviously lower than other groups (P < 0.05). Conclusion Jinshuixian combined with radiotherapy has the ability of blocking proliferation and inhibiting the expression of HIF-1α and VEGF.
